Tittle: Eukaryotic production and characterization of multi-protein complexes: tools and examples

Characterization of multi-protein complexes and their reconstitution from recombinant sources are becoming increasingly important as the focus of molecular and structural biology progresses from analysis of individual proteins to that of multi-subunit assemblies. Based on case studies, we will discuss our latest developments to facilitate the production of recombinant mid-size complexes (3 to 9  protein subunits): (i) multi-host plasmids libraries for expression/co-expression screening in insect or mammalian cells and (ii) streamlined procedures to assemble multi-gene expression constructs from synthetic biology approaches. We also will illustrate strategies to generate mammalian stable cell lines as well as the use of a CrispR/Cas9-based approach to affinity-tag large macromolecular assemblies (10 subunits and more) and facilitate their purification from endogenous source.

Speaker: Adrien Trolet, Serendip, Strasbourg, France

Tittle: A versatile plant-derived drug delivery platform based on phytoviruses.

In recent years, plant-made pharmaceuticals have emerged as a promising approach to biopharmaceutical production, leveraging the natural efficiency of plants as bioreactors for synthesizing complex therapeutic molecules. Since the first recombinant hormone was produced in tobacco in 1986, numerous companies have successfully developed a diverse range of molecules—from monoclonal antibodies to virus-like particles—many of which have reached clinical trials and even secured market approval.

Building on the cost-effectiveness, scalability, and strong safety profile of plant-based protein manufacturing, Serendip Innovations, a French biotech company, is pioneering a next-generation immunization platform aimed at enhancing the bioavailability and stability of active pharmaceutical ingredients, such as proteins and peptides.

This presentation will explore how this innovative production system could revolutionize the biopharmaceutical industry and improve global access to essential medicines. We will also showcase case studies of successful plant-based production systems, their applications in treating various diseases, and the regulatory challenges shaping this rapidly evolving field.
